In isolated primary murine neonatal microglia and the murine BV-2 microglia cell line, exogenous LPA induces a pro-inflammatory phenotype, and many of these inflammatory reactions are attenuated in the presence of LPA5 antagonists. When BV-2 microglia are polarized with LPS, they increase ATX transcription and LPA secretion, along with the release of proinflammatory cytokines.
To get an indication about the effects of LPS on ATX expression, we performed Western blot experiments. LPS treatment increased the expression of ATX at 8 and 24 h.
The antibody was used at 1:500 dilution in 5% BSA in PBS (overnight incubation of the plots at +4°C). A clear band was observed at the expected/given molecular weight. Even though some antibodies from Santa Cruz do not work well/give a lot of unspecific binding, we had no problems with this one, giving a "clean" background. I highly recommend long washes before developing the plot. We usually wash for 1.5-2 hours (changing the wash buffer every 20 minutes). All and all, an unexpectedly good antibody which I definitely recommend for ATX detection.
