Great quality host cells in the form of bacterial cells are highly required for cloning, as we have to transfer the vector into these cells, and then they have to multiply for growth. For this purpose, Invitrogen competent cells are a great option, which I use on a regular basis for cloning various genes. These competent cells show excellent growth over the agar plates, but one thing we have to keep in mind is that we have to make aliquots of these cells, and use it immediately after thawing as per the required quantity. Regular and frequent thawing doesn't give good results, so you have to make aliquots and store accordingly at the appropriate temperatures.
