We routinely use the gentleMACS™ Octo Dissociator with Heaters for brain tissue dissociation and subsequent isolation of astrocyte and microglia cells from mouse brains. The equipment requires the use of specific C tubes (catalog number: 130-093-237), into which tissue pieces are placed in the enzymatic solution. The C tubes are attached upside down onto the sleeve of the gentleMACS™ Octo Dissociator; therefore, it is important to tightly close the C tube before attaching it to the instrument. The instrument provides multiple programs for tissue dissociation, which should be chosen based on the tissue type and protocol used. In our lab, two programs are routinely used (37C_ABDK-01 for the whole mouse brain and 37C_ABDK-02 for dissected mouse brain regions). It takes 30 minutes to run these programs. Both programs work well, and the dissociated tissue provides a high yield of isolated astrocytes following the Isolation and Cultivation of Astrocytes from Adult Mouse Brain Miltenyi Biotec protocol. The instrument is benchtop-sized and can process eight samples simultaneously. Another great feature of the equipment is that all eight positions can be operated autonomously. I highly recommend this instrument as it saves plenty of time and effort in obtaining single-cell suspensions.
