The MACSQuant Tyto is equipped with three lasers, which allows cell sorting with up to 10 parameters. Since the actual sorting process takes place completely within the Tyto Cartridge, there is no risk of losing precious sample material and therefore the sorting process is entirely operator-free. With its extremely compact design, the MACSQuant Tyto is a true benchtop instrument that perfectly fits into a typical lab environment.
The heart of the system is the MACSQuant Tyto Cartridge, a single-use disposable carrying the actual sorting microchip. Unlike in conventional droplet sorters, no fluidic tubing is present in the instrument itself. Additionally, cells sorted by the MACSQuant Tyto do not experience high sorting pressure, charge, or decompression. Instead, a very mild and constant filtered air pressure (< 3 psi) drives the cells through the microchip for sorting, and subsequently either into the positive or negative collection chamber (two-way sorting). The lack of fluidic tubing in the system means that fluidics start-up and laborious cleaning procedures are no longer required, saving operator-time and reducing costs. Furthermore, sorting temperature can be cooled down to 4 °C, providing optimal conditions to retain highest cell viabilities and functionalities.
