Murine TNF-α Mini TMB ELISA Development Kit
The goal of our project was to investigate the role of the ATX/LPA/LPARs axis under inflammatory conditions.
In order to study the effects of autotaxin and LPAR5 inhibition during systemic endotoxemia, we used different ELISAs to evaluate possible changes in the levels of proinflammatory cytokines in the serum of mice. Mice were injected i.p. with DMSO, LPS (5 mg/kg) plus DMSO, with or without PF8380 (‘PF’; ATX inhibitor) or AS2717638 (‘AS’; LPAR5 inhibitor). After 24 h, the animals were sacrificed, and the blood was collected and processed to obtain serum. Serum was diluted at 1:10 for further analysis.
The TNFa ELISA was mainly used according to manufacturer's instructions. Only difference is that we decided to incubate the samples overnight at 4°C.
The ELISA is cheap and easy to use and the only caveat is that the plates are not pre-coated.
