Assessment of Atp Levels Using Celltiter-Glo® 3D Cell Viability Assay

The CellTiter-Glo 2.0 Luminescent Cell Viability Assay (Promega, UK) measures total ATP levels produced by metabolically active cells. I used this product to assess ATP levels in chondrocytes treated with extracellular vesicles (EVs). Luminescence measurements were performed on day 5 of cell culture, based on 100 μl of equilibrated CellTiter-Glo 2.0 reagent. Luminescence was measured 10 min afterward, using the Spark multimode microplate reader (Tecan, Switzerland) and the Sparkcontrol method editor software (Tecan).
