For experiments, age-matched embryos were obtained via egg lay with gravid adults, then raised to the L4 stage at 15°C. Worms were then picked into 96-well plates for Resipher (Lucid Scientific) analysis at a density of 20 worms/well in a volume of 100 µL containing OP50 E. coli bacteria for food and vehicle (0.5% DMSO), 5 µM, or 20 µM DCCD (ATP synthase inhibitor). OCR was measured continuously for 24h, following which lethality was visually assessed under the microscope after a harsh touch with a platinum wire. Panel b shows OCR at 4h time intervals throughout the measurement, normalized per worm. Each data point shown is the average of 4 technical replicates. Experiments were carried out in 4 biological replicates. Bars and error bars show the mean and SEM, respectively. No significant differences were observed between ploidy groups in a two-way ANOVA with Bonferroni-adjusted post testing for individual comparisons. Panel c shows the lethality following DCCD exposure in the Resipher plate. No significant differences were observed between ploidy groups.
