We acquired this microscope to fulfill several purposes, including cell counting and climate controlled experiments using both blood leukocytes (neutrophils, monocytes) and epithelial cells. Specific objectives included assessing caspase activity over time following viral infection, quantifying cell death, and counting cells after migration through transwells to determine yield. Counting adherent epithelial cells stained with a nuclear dye works fine, but counting of non adherent cells such as monocytes was never close to the accuracy of traditional hemocytometer. I invested considerable amounts of time troubleshooting with scientists and technical support to correct issues with the environmental control and software, thus creating a huge activation energy just to obtain my basic endpoints. I can see the great potential with this machine but it required a fair amount of frustration in the initial set up. However, Molecular Devices does offer tremendous customer support and their personnel are completely committed to helping the customer with any problems that arise.
