HPLC UV VIS Detector

Because of their relative ease-of use and high sensitivity, UV VIS detectors are the most common type of detector used for high pressure liquid chromatography (HPLC). HPLC UV VIS detectors operate by passing visible and UV light through a sample in a flow cell, then measuring the absorption of the different wavelengths that pass through the cell. The amount of light absorbed provides information on the properties of the sample of interest. There are two types of UV VIS detectors: tunable detectors and photodiode array (PDA) detectors. Tunable detectors measure absorbance at one or more discrete wavelengths—most commonly 254 nm, while PDA detectors measure a spectrum of wavelengths simultaneously, thus increasing sensitivity and analysis speed in cases where measurement of multiple wavelengths is required. Other factors to consider when choosing an HPLC UV VIS detector is the wavelength range that can be captured, ease of operation, and its compatibility with the HPLC system to be used.
