A Luminescent-based Assay for Quantification of Glutathione (GSH) in Cells

Intracellular Glutathione content was measured using GSH-Glo™ Glutathione Assay Kit according to the manufacturer’s protocol. Microglia were seeded in black clear bottom 96-well plates at a density of 5x103 cells per well and incubated in serum-free media containing LPA 1 µM for indicated time periods. Then, GSH-Glo reagent was added to the plate and incubated for 30 min. Finally, after addition of luciferin detection reagent, luminescence was measured to quantify the glutathione content.

The kit is easy to use and results can obtained after 30 minutes. We observed a variation between replicates and it is recommended to perform multiple experiments using many replicates.
