Adenomatous polyposis coli (APC) is a large protein of ~312 kDa that functions as a tumor suppressor. It is a critical scaffolding protein of the beta-catenin destruction complex in the WNT pathway. Immunoblotting APC from cell extracts can be challenging because of its large size. I use 3 to 8% Tris-Acetate gel for this purpose and it can even resolve post-translationally modified APC. I typically use this gel for resolving proteins >250 kDa.
